Significance |
Firefly luciferase is commonly used as a reporter for gene regulation and function in a wide variety of research areas. The enzyme catalyzes conversion of D-luciferin to oxyluciferin, a reaction that requires ATP and oxygen and results in the emission of light centered at 560 nm. The SpectraMax®Glo Steady Luc™ Reporter Assay uses a proprietary formulation to modify the enzymatic reaction for a longer signal half-life of about 3 hours.
Optimized settings for assay detection using the SpectraMax M5 Multi-Mode Microplate Reader are provided in the protocol. Similar settings will work on other plate readers with luminescence detection mode. The instrument is set up to detect light of all wavelengths (no wavelength selection) with an integration time of 1000 msec (1 sec). |
